I bought this skill, but the game won't tell me whether the threads I've found belong to men or women. Am I doing something wrong?

Nope! Just because you have the skill, doesn't mean that you can automatically tell which threads are what. You have the ability to sometimes tell which are which.
It's kind of like the lotto - you can't win without buying a ticket, but buying a ticket doesn't garuntee you winning.

Whether these skills have effect or not, also depends on your equipement. Try using equipement that enhances your smarts.

Does anyone have a feel for when the determination is made for what you can find. For instance, if I start a case, then go to my agency and load up on smart items and go to the crime scene, will this increase my chances, or do I need to have the smart items equipped when I start the case.

Yes, it definitely increases your chances if you put on smarts right before you go to the crime scene.

I understand that max is asking *when* physical evidence is determined. If the dice is thrown when you click on start new case, then changing equipement before going to the crime scene won't make a difference.
I actually have no idea when this happens, it feels more natural if it is just before going to the crime scene, but only the creator knows :)
