do you need to be in the same city that you write a script for?
especially for testing? Or does that not matter...
Please advice..

You can write your mystery while in any city, but to test it you will have to travel to the city where the story is set.

It's a good idea on your first mystery you write to set it in the city you are based in as you will take a while to get to know the system for opening questions etc.
