Hi Nick - the specific questions you can ask have mostly to do with villain hunts, which are a relatively new feature of the game. Unfortunately villain hunting is only available to players who are subscribed and who have a great deal more experience than you do. For now there isn't anything you can ask the townsfolk that will alter your game in any way.
If you just want them to say ANYTHING other than "I don't know anything about that" for fun, try asking about Big Lucy or Cairo. Won't help you in any way, but at least you'll have the satisfaction of making them stop repeating themselves.
